用途:
依ASTMD1148制造、該機(jī)作為模擬陽光的紫外線輻射及熱度,試件于機(jī)臺(tái)內(nèi)經(jīng)紫外線照射與溫度作用、經(jīng)過一段時(shí)間后觀察試件耐黃變的程度,用灰色標(biāo)作參考,判定其黃變的等級(jí),機(jī)臺(tái)附件齊全亦可當(dāng)老化試驗(yàn)機(jī)及烘箱使用。

使用注意事項(xiàng):
1、電源設(shè)備:應(yīng)在供電線路中裝有超負(fù)荷的保險(xiǎn)絲裝置,供此箱特種,并具有良好接地裝置。
2、試料準(zhǔn)備:將試品逐一編號(hào)后,將試品放置于試品轉(zhuǎn)盤上,彼此以不相互接觸和碰撞為宜。
3、待一切準(zhǔn)備就緒,即關(guān)上箱門。
4、關(guān)閉控制系統(tǒng)各個(gè)開關(guān),將電源插撲插上打開電源開關(guān)轉(zhuǎn)盤即自動(dòng)轉(zhuǎn)動(dòng),同時(shí)控制系統(tǒng)的電源指示燈即亮,若須升溫必須打開開關(guān)(即高溫開關(guān)和低溫開關(guān)),然后調(diào)節(jié)全自動(dòng)控制系統(tǒng)
5、儀器外殼必須有效接地,以保證使用安全。

按理化性質(zhì)和生物學(xué)功能,可將其分為IgM、IgG、IgA、IgE、IgD五類。IgM抗體是免疫應(yīng)答中首先分泌的抗體。它們在與抗原結(jié)合后啟動(dòng)補(bǔ)體的級(jí)聯(lián)反應(yīng)。它們還把入侵者相互連接起來,聚成一堆便于巨噬細(xì)胞的吞噬;IgG抗體激活補(bǔ)體,中和多種毒素。IgG持續(xù)的時(shí)間長,是唯一能在母親妊娠期穿過胎盤保護(hù)胎兒的抗體。他們還從乳腺分泌進(jìn)入初乳,使新生兒得到保護(hù);IgA抗體進(jìn)入身體的黏膜表面,包括呼吸、消化、生殖等管道的黏膜,中和感染因子。還可以通過母乳的初乳把這種抗體輸送到新生兒的消化道黏膜中,是在母乳中含量最多,最為重要的一類抗體;IgE抗體的尾部與嗜堿細(xì)胞、肥大細(xì)胞的細(xì)胞膜結(jié)合。當(dāng)抗體與抗原結(jié)合后,嗜堿細(xì)胞與肥大細(xì)胞釋放組織胺一類物質(zhì)促進(jìn)炎癥的發(fā)展。這也是引發(fā)速發(fā)型過敏反應(yīng)的抗體;IgD抗體的作用還不太清楚。它們主要出現(xiàn)在成熟的B淋巴細(xì)胞表面上,可能與B細(xì)胞的分化有關(guān)。(IgD于1995年從人骨髓瘤蛋白中發(fā)現(xiàn),分子量為175kD,主要由扁桃體、脾等處漿細(xì)胞產(chǎn)生,人血清中IgD濃度為3~40μg/ml,不到血清總Ig的1%,在個(gè)體發(fā)育中合成較晚。IgD鉸鏈區(qū)很長,且對(duì)蛋白酶水解敏感,因此IgD半衰期很短,僅2.8天。血清中IgD確切的免疫功能尚不清楚。在B細(xì)胞分化到成熟B細(xì)胞階段,除了表達(dá)SmIgD,抗原刺激后表現(xiàn)為免疫耐受。成熟B細(xì)胞活化后或者活化后或者變成記憶B細(xì)胞時(shí),SmIgD逐漸消失。

抗體是具有4條多肽鏈的對(duì)稱結(jié)構(gòu),其中2條較長、相對(duì)分子量較大的相同的重鏈(H鏈);2條較短、相對(duì)分子量較小的相同的輕鏈(L鏈)。鏈間由二硫鍵和非共價(jià)鍵聯(lián)結(jié)形成一個(gè)由4條多肽鏈構(gòu)成的單體分子。輕鏈有κ和λ兩種,重鏈有μ、δ、γ、ε和α五種。 整個(gè)抗體分子可分為恒定區(qū)和可變區(qū)兩部分。在給定的物種中,不同抗體分子的恒定區(qū)都具有相同的或幾乎相同的氨基酸序列。可變區(qū)位于"Y"的兩臂末端。在可變區(qū)內(nèi)有一小部分氨基酸殘基變化特別強(qiáng)烈,這些氨基酸的殘基組成和排列順序更易發(fā)生變異區(qū)域稱高變區(qū)。高變區(qū)位于分子表面,最多由17個(gè)氨基酸殘基構(gòu)成,少則只有2 ~ 3個(gè)。高變區(qū)氨基酸序列決定了該抗體結(jié)合抗原抗原的特異性。一個(gè)抗體分子上的兩個(gè)抗原結(jié)合部位是相同的,位于兩臂末端稱抗原結(jié)合片段(antigen-binding fragment, Fab)。"Y"的柄部稱結(jié)晶片段(crystalline fragment,FC),糖結(jié)合在FC 上。
